Study on antioxidant activity of Artocarpus altilis L. leaves

Abstract

The aim of this study was to determine the in vitro and in vivo antioxidant capacities of Artocarpus altilis L. leaves. Besides, the main composition of A.altilis leaves was also identified. The in vitro antioxidant activityof water extract of Artocarpus altilis L. leaves was evaluated by using high performance liquidchromatography – electron spin resonance (HPLC-ESR) technique. The in vivo antioxidant abilities of groundpower and water extract of A.altilis leaves were determined by using gene modified Drosophila melanogaster (GMR-GAL4/UAS-hDuox2) model with rough-eye phenotypes. HPLC-ESR method was carried out byinjecting of 4 mg/ml water extract into a TSK-gel-size exclusion column (TOSOH, G3000PW, inner diameterof 7.5 mm, and length of 30 cm) and monitoring ESR signal intensity of radicals adduct of 5,5-dimethyl-1-pyrroline N-oxide (DMPO/O\(_2\)). This product was then measured at 254 nm wavelength. The results revealedthat leaf water extract of A. altilis contains aromatic compounds, glucose, sucrose, amino acid, and fatty acid.HPLC-ESR analysis showed that the concentration of product of reaction of DMPO with O\(_2\) (DMPO/O\(_2\))decreased 20% and the free radical absorption reduced from 0.2 to approximately 0.15. The ground power andthe water extract of A. altilis were carried out in vivo at various concentration including 0% (considered as thecontrol sample), 5%, 10% and 20%. In addition, the application of 20% ground power and aqueous extract torough-eye phenotypes resulted in the recovery to normal phenotypes at the percentage of 75.7% and 84.25%,respectively. In conclusion, the in vitro and in vivo experiments indicated that both ground power and waterextract from leaves of A. altilis possess effective antioxidative abilities.
