AFLP analysis of genetic diversity among populations of black tiger shrimp (Penaeus monodon) collected from coastal regions of Vietnam

Abstract

Black tiger shrimp (Penaeus monodon) is one of the most important aquaculture species in Vietnam and contributes significantly to the country economic development by its enormous commercial value. The strategy for shrimp industry development is to achieve stable, sustainable production with minimal negative environmental impact. A cornerstone for this strategy is the development of domesticated stocks of P. monodon and rational breeding programs for improved survival and growth. The study of genetic diversity and search for molecular markers that link with economically important traits such disease resistance, reproductive efficiency and product quality is a key issue for P. monodon breeding programs. In this study, the P. monodon samples from different geographical locations of Vietnam (BTB, NTB and NB) were collected in order to analyze genetic diversity of the wild populations by AFLP technique using AFLP® Mapping Kit (Applied Biosystems). Total DNA from the samples were extracted, purified and cut with EcoRI and MseI, then was linked with EcoRI and MseI adaptors available in the Kit. The adaptor linked DNA fragments were amplified by PCR using preselective primers and selective primers MseI-CTT, EcoRI-ACC JOE in which JOE is fluorescent dye. AFLP products were analyzed by GeneMapper® Software Version 4.1 AFLP® System Analysis to determine the number of alleles and the polymorphism of the three investigated populations. The alleles obtained from the BTB, NTB and NB populations ranged from 32 to 76, 29 to 64 and 38 to 39, respectively. The coincident alleles in the NTB population were 4, in the NB population were 8 and no coincident alleles detected among individuals within the BTB population. The subsequent analysis showed that, there was no coincident allele among the three investigated populations. From the results, we can primarily conclude that, there is a high genetic variability among wild black tiger shrimp populations of Viet Nam. Our results provided scientific basis for further research in order to reserve and exploit indigenous genetic resource for selective breeding programs.
